If he doesn't ask the question, this will stay in his mind forever. It will bother him for a little while and then it will go away, only to make its presence every now and then. I say go ahead and ask.
You didn't venture the third possibility. She teslls the truth; which has two more outcomes:
1) She has slept with only one (ex).
2) She has slept with more than 1.
