You don't need to have any experience if your your employer advertised your job description which requires master degree. You even can have 0 years of experience.

Also it doesn't matter how your h1b was applied (Bachelor quota or masters quota). Green card is for future employement and it has nothing to do with your past history (how and where you got your h1b or any other visa).
