based on my own experience, no. your bro would need a canadian visitor visa. can try applying for on in one of the canadian consulates in the us. good luck.
If he has the multiple entry to the US, yes, he can enter the US, but first he has to get the visa for Canada to enter that country.

I believe "NO". People who used to come on B1/B2 visa, used to save bucks by renewing their 6 months visa (which they get at airport) by going to CAN/MAX. Now this "KALE" president changed the rule so inorder to renew your visa (I know you have 5 year multiple but to get 6 month visa while entering US), you must travel the countries other then North and South America. Just talk with other experts also.
