After you obtain your US citizenship, you can relinquish your Nepali citizenship by following the link below:

Why do you want to give up your Nepali Citizenship? It might be useful if Nepal passes the dual citizenship bill. That way you don't need to apply for a visa for a Nepal visit.

Let me count how many Nepali citizens have given up their Nepali citizens after being US Citizens.
Answer: may be none to 1-2 ( need data)

You need your Nepali citizenship every wake of your life if you have to deal in Nepal for many reasons.
If you have any property specially land: without citizenship you can not do any thing.

My answer is : NO , you do not have to give up.
