I am in OPT status right now. My opt expires on Aug. 2016. My Employer is going to file H1B in 2016 April. As all of us know, H1B is not 100% sure. So i am thinking to apply TPS for backup. what will happen if i apply for TPS now and later in April apply for H1B too? Does my TPS will have negative impact on my H1b?

No, at this point I would suggest you to apply TPS, and also TPS EAD both. if you have financial problem file with waiver too. There is no harm at all.
